Key information
Overview
AU FOOD aims to acquire high‑throughput, high‑resolution MS equipment optimized for proteomics runs, due to new projects on alternative protein sources (e.g., single‑cell protein). AU FOOD currently operates a TimTof Pro (2nd generation dual TIMS) system, which is intended to be upgraded to an HT (4th generation TIMS XR). This upgrade will deliver higher analysis speed (MS/MS speed ~300 Hz PASEF) and increased ion capacity, enabling the analysis of larger peptide loads without saturation, as well as a significantly improved quantitative dynamic range achieved through the introduction of Advanced Digitizer Technology (ADT). There is a need to identify more proteins in complex samples. The upgrade is expected to result in approximately 70–76% more identified parent ions/precursors. We also seek improved reproducibility in the quantification of parent ions/precursors, which will improve by more than twofold (>2×) with the HT system. The HT system enables more than a halving of the run time per sample (from 30–60 minutes with the current system to 5–30 minutes with HT), allowing for well over a doubling of the number of samples processed per day (from currently 10 samples per day to more than 50 samples per day). Furthermore, HT provides improved dia‑PASEF performance in terms of scalability and reproducibility, enabling large‑scale proteomics, as well as an improved injection system with reduced maintenance and increased runtime. The upgrade also includes improved software optimized with TIMScore™, TIMS DIA NN, and ProteoScape™. Bruker is the only supplier able to provide this upgrade, carried out by certified technicians and followed by a warranty on the upgraded system. Given the high cost of high‑resolution MS equipment from all suppliers, upgrading the existing system has been identified as a cost‑effective way to obtain equipment that meets the highest current standards.
